چکیده

Fumarate hydratase (FH), encoded by the FH gene, is an enzyme which catalyses conversion of fumarate to L-malate as part tricarboxylic acid cycle. Biallelic germline mutations in result fumaric aciduria, a metabolic disorder resulting severe neurological and developmental abnormalities. Heterozygous hereditary leiomyomatosis renal cell carcinoma, cancer predisposition syndrome. deficiency has multiple oncogenic mechanisms including through promotion aerobic glycolysis, induction pseudohypoxia, post-translational protein modification impairment DNA damage repair homologous recombination. FH-deficient neoplasms can present with characteristic morphological features that raise suspicion for alterations also frequently demonstrate loss immunoreactivity intracellular accumulation 2-succinocysteine, detected immunohistochemistry.

Gene of the month: SMARCB1

SMARCB1 is the core subunit of the SWI/sucrose non-fermenting ATP-dependent chromatin remodelling complex located on the long arm of chromosome 22 (22q11.2). Since discovering genetic alterations of the SMARCB1 gene in malignant rhabdoid tumours, the family of tumours harbouring loss of SMARCB1 expression has been steadily expanding. In this review, we give a general overview of SMARCB1, its ro...

Gene of the month: HGF.

Hepatocyte growth factor (HGF) is a multifunctional cytokine with important roles in cell proliferation, survival, motility and morphogenesis. Secreted by cells of mesenchymal origin, HGF is the specific ligand for the tyrosine-kinase receptor c-MET (cellular mesenchymal-epithelial transition), also called MET, which is expressed in different types of epithelial, endothelial and haematopoietic ...

CYS127S (FH-Kairouan) and D245N (FH-Tozeur) mutations in the LDL receptor gene in Tunisian families with familial hypercholesterolaemia.

Familial hypercholesterolaemia (FH, MIM 143890) is a dominantly inherited metabolic disorder caused by various mutations of the LDL receptor (LDL-R) gene causing delayed clearance of serum low density lipoproteins (LDL). The prevalence of homozygous FH has been estimated to be 1 in a million in European and North American populations. In central and southern Tunisia, the prevalence of homozygou...
